The invention relates to a synthetic method for N-t-butyloxycarboryl-N'-(pentamethyldihydrobenzofuran-5-sulfonyl)-L-arginine, mainly solving the technical problem of lack of synthetic methods for theN-t-butyloxycarboryl-N'-(pentamethyldihydrobenzofuran-5-sulfonyl)-L-arginine. The synthetic method includes the following steps of dropwise adding an acetone solution of 2,2,4,6,7-pentamethyldihydrobenzofuran-5-sulfonyl chloride into a water solution of N-t-butyloxycarboryl-L-arginine monohydrochloride monohydrate and sodium hydroxide under ice water bath, performing substitution reaction at a lowtemperature, adjusting the pH to 2 by hydrochloric acid liquor, performing extraction by ethyl acetate, performing washing and concentration, and crystalizing a crude product to obtain a target product 1. The N-t-butyloxycarboryl-N'-(pentamethyldihydrobenzofuran-5-sulfonyl)-L-arginine serving as a non-natural amino acid derivative is mainly applied to synthesis of polypeptide.
