Ferric ion concentration-controlled aerobic photo-oxidation of benzylic C–H bond with high selectivity and conversion
A Fe(III)-promoted highly selective photo-oxidation of benzylic C–H bond delivering relative carbonyl products is reported. By altering the concentration of ferric salt, methylarenes can be selectively oxidized under UV irradiation to furnish aromatic aldehydes or acids, respectively. By this protocol, the oxidation of ethylarenes provides the corresponding acetophenones. The reaction is inferred to involve divergent pathways in different concentrations of catalyst for the alternative selectivity between aldehydes and aicds. The reusable catalyst, high conversion and selectivity make this oxidation a green and economic protocol for the synthesis of aromatic carbonyl compounds.

Preparation process of high-purity 3,5-bis(trifluoromethyl)benzyl alcohol
The invention discloses a preparation method of high-purity 3,5-bis(trifluoromethyl)benzyl alcohol, and belongs to the technical field of organic synthesis. The method comprises the steps: reacting a Grignard reagent generated by reaction of 3,5-bis(trifluoromethyl)bromobenzene and magnesium metal with a reagent capable of introducing an aldehyde group to obtain 3,5-bis(trifluoromethyl)benzaldehyde, and then reducing 3,5-bis(trifluoromethyl)benzaldehyde into 3,5-bis(trifluoromethyl)benzyl alcohol through sodium borohydride. The purity of the product obtained by the technical route is more than 99.5%, the maximum single impurity is not more than 0.1%, and the requirement on high-purity 3,5-bis(trifluoromethyl)benzyl alcohol in the market is met. Raw materials required by the process are easy to obtain, reaction conditions are mild, safety is high, production cost is low, and the method is suitable for industrialization.

Asymmetric Organocatalysis and Continuous Chemistry for an Efficient and Cost-Competitive Process to Pregabalin
Herein, we present the scale up development of an innovative synthetic process to pregabalin. The process is underpinned by two enabling technologies critical to its success; continuous chemistry allowed a safe and clean production of nitroalkene, and asymmetric organocatalysis gave access to the chiral intermediate in an enantioenriched form. Crucial to the success of the process was the careful development of a continuous process to nitroalkene and optimization of the organocatalyst and of the reaction conditions to attain remarkably high turn-over frequency in the catalytic asymmetric reaction. Successful recycle of the organocatalysts was also developed in order to achieve a cost-competitive process.

Rhodium-catalyzed reductive carbonylation of aryl iodides to arylaldehydes with syngas
The reductive carbonylation of aryl iodides to aryl aldehydes possesses broad application prospects. We present an efficient and facile Rh-based catalytic system composed of the commercially available Rh salt RhCl3·3H2O, PPh3 as phosphine ligand, and Et3N as the base, for the synthesis of arylaldehydes via the reductive carbonylation of aryl iodides with CO and H2 under relatively mild conditions with a broad substrate range affording the products in good to excellent yields. Systematic investigations were carried out to study the experimental parameters. We explored the optimal ratio of Rh salt and PPh3 ligand, substrate scope, carbonyl source and hydrogen source, and the reaction mechanism. Particularly, a scaled-up experiment indicated that the catalytic method could find valuable applications in industrial productions. The low gas pressure, cheap ligand and low metal dosage could significantly improve the practicability in both chemical researches and industrial applications.

Rhodium-Catalyzed Formylation of Aryl Halides with CO2 and H2
The reductive formylation of aryl iodides/bromides to aryl aldehydes using CO2/H2 is presented for the first time. It was realized over a catalytic system composed of RhI3 or RhI3/Pd(dppp)Cl2 (dppp = 1,3-bis(diphenyphosphino)propane) and PPh3 in the presence of Ac2O/Et3N at 100 °C, affording aromatic aldehydes in good to excellent yields, together with good functional-group tolerance and broad substrate scope. The reaction proceeds through three cascade steps, involving HCOOH formation, CO release, and formylation of aryl halides.

Bifunctional Biphenyl-2-ylphosphine Ligand Enables Tandem Gold-Catalyzed Propargylation of Aldehyde and Unexpected Cycloisomerization
Despite extensive studies in gold catalysis, σ-allenylgold species have not been invoked as catalytic intermediates and their reactivities not studied. This work reports for the first time they are generated in situ and undergo nucleophilic addition to activated aldehydes in a bifunctional phosphine ligand-enabled gold catalysis. This development reveals a broad range of opportunities to achieve propargylic C-H functionalization for the first time under catalytic and mild conditions. The homopropargylic alcohols generated undergo ligand-enabled cycloisomerizations involving an unexpected silyl migration.

Syntheses and biological evaluation of 1,2,3-triazole and 1,3,4-oxadiazole derivatives of imatinib
Three novel series of 1,2,3-triazole and 1,3,4-oxadiazole derivatives of imatinib were prepared and evaluated in vitro for their cytostatic effects against a human chronic myeloid leukemia (K562), acute myeloid leukemia (HL60), and human leukemia stem-like cell line (KG1a). The structure-activity relationship was analyzed by determining the inhibitory rate of each imatinib analog. Benzene and piperazine rings were necessary groups in these compounds for maintaining inhibitory activities against the K562 and HL60 cell lines. Introducing a trifluoromethyl group significantly enhanced the potency of the compounds against these two cell lines. Surprisingly, some compounds showed significant inhibitory activities against KG1a cells without inhibiting common leukemia cell lines (K562 and HL60). These findings suggest that these compounds are able to inhibit leukemia stem-like cells.

Preparation and application of novel Swern reagent
The invention discloses 4-(2-(2-methyl sulfoxide)ethyl)-4-nitrobenzene)morpholine shown in the formula (I) and preparation and application thereof. A preparation method includes the steps that 2-(2-chlorine-5 nitro)phenethyl alcohol shown in the formula (II) and morpholine are mixed to prepare 2-(2-morpholine-5-nitrobenzene)ethanol shown in the formula (III); bis(trichloromethyl)carbonate ester, a sodium methyl mercaptide aqueous solution and an aqueous hydrogen peroxide solution are sequentially added dropwise to 2-(2-morpholine-5-nitrobenzene)ethanol shown in the formula (III), and finally 4-(2-methyl sulfoxide)ethyl)-4-nitrobenzene)morpholine is prepared. According to the application of 4-(2-methyl sulfoxide)ethyl)-4-nitrobenzene)morpholine, the obtained Swern reagent reacts with an alcohol compound shown in the formula (IV), and aldehyde or ketone is prepared after after-treatment. The defects of an existing Swern oxidation method are overcome, generation of a stink byproduct dimethyl sulfide and toxic carbon monoxide is avoided from the source, the reaction temperature is increased to be -30 DEG C to 0 DEG C, and an odorless byproduct novel sulfur ether can be recycled and reused. Selective oxidation of amines to aldehydes or imines using laccase-mediated bio-oxidation
An efficient and practical chemo-enzymatic aerobic oxidation in water of benzylamines to obtain aldehydes or imines is described. Laccase from Trametes versicolor was chosen as biocatalyst, and TEMPO (radical 2,2,6,6-tetramethylpiperidine 1-oxyl) as mediator. A study on the pH dependence of the aqueous medium allowed us to realise a fine tuning on product selectivity. Under our optimized reaction conditions, the bio-oxidation of a series of primary, secondary and cyclic amines has been achieved.

Laccase-Mediator System for Alcohol Oxidation to Carbonyls or Carboxylic Acids: Toward a Sustainable Synthesis of Profens
By combining two green and efficient catalysts, such as the commercially available enzyme laccase from Trametes versicolor and the stable free radical 2,2,6,6-tetramethylpiperidine-N-oxyl (TEMPO), the oxidation in water of some primary alcohols to the corresponding carboxylic acids or aldehydes and of selected secondary alcohols to ketones can be accomplished. The range of applicability of bio-oxidation is widened by applying the optimized protocol to the oxidation of enantiomerically pure 2-arylpropanols (profenols) into the corresponding 2-arylpropionic acids (profens), in high yields and with complete retention of configuration.

Practical Cu(OAc)2/TEMPO-catalyzed selective aerobic alcohol oxidation under ambient conditions in aqueous acetonitrile
We reported a ligand-and additive-free Cu(OAc)2/TEMPO catalyst system that enables efficient and selective aerobic oxidation of a broad range of primary and secondary benzylic alcohols, primary and secondary 1-heteroaryl alcohols, cinnamyl alcohols, and aliphatic alcohols to the corresponding aldehydes and ketones. This ambient temperature oxidation protocol is of practical features like aqueous acetonitrile as solvent, ambient air as the terminal oxidant, and low catalyst loading, presenting a potential value in terms of both economical and environmental considerations. Iron chloride/4-acetamido-TEMPO/sodium nitrite-catalyzed aerobic oxidation of primary alcohols to the aldehydes
A variety of 4-substituted 2,2,6,6-tetramethylpiperidyl-1-oxy (TEMPO) derivatives has been screened for their ability in the oxidation of primary alcohols to the aldehydes with dioxygen under mild conditions. An evaluation of the efficiency of these 4-substituted TEMPO derivatives in the alcohol oxidation may allow an insight into the effect of the structural variations of TEMPO on the oxidation of alcohols, which should facilitate catalyst design and screening efforts. Development of safe and scalable continuous-flow methods for palladium-catalyzed aerobic oxidation reactions
The synthetic scope and utility of Pd-catalyzed aerobic oxidation reactions has advanced significantly over the past decade, and these reactions have the potential to address important green-chemistry challenges in the pharmaceutical industry. This potential has not been realized, however, because safety concerns and process constraints hinder large-scale applications of this chemistry. These limitations are addressed by the development of a continuous-flow tube reactor, which has been demonstrated on several scales in the aerobic oxidation of alcohols. Use of a dilute oxygen gas source (8% O2 in N 2) ensures that the oxygen/organic mixture never enters the explosive regime, and efficient gas-liquid mixing in the reactor minimizes decomposition of the homogeneous catalyst into inactive Pd metal. A practical non-cryogenic process for the selective functionalization of bromoaryls
A selective and practical bromine-metal exchange process under non-cryogenic conditions was developed by a simple modification of an existing protocol. By directly adding an alkyl lithium RLi reagent to a solution of a bromoaryl substrate ArBr and an alkylmagnesium reagent RMgX, a lithium triarylmagnesiate Ar3MgLi complex formed that allowed for various types of functionalization and more elaborate cross-coupling reactions. The simplicity and improved safety of the method represent a significant improvement over current state of the art that uses lithium trialkylmagnesiate R3MgLi complexes, and is especially advantageous for large-scale synthesis.

TEMPO/HCl/NaNO2 catalyst: A transition-metal-free approach to efficient aerobic oxidation of alcohols to aldehydes and ketones under mild conditions
Hydrochloric acid, a very inexpensive and readily available inorganic acid, has been found to cooperate exquisitely with NaNO2/TEMPO in catalyzing the molecular-oxygen-driven oxidation of a broad range of alcohol substrates to the corresponding aldehydes and ketones. This transition-metal- free catalytic oxidative conversion is novel and represents an interesting alternative route to the corresponding carbonyl compounds to the metal-catalyzed aerobic oxidation of alcohols.The reaction is highly selective with respect to the desired product when carried out at room temperature in air at atmospheric pressure. Notably, the use of very inexpensive NaNO2 and HCl in combination with TEMPO for this highly selective aerobic oxidation of alcohols in air at ambient temperaturemakes the reaction operationally and economically very attractive. The results of mechanistic studies, performed with the aid of electrospray ionization mass spectrometry (ESI-MS), are presented and discussed. TEMPO, TEMPOH, and TEMPO+ were observed in the redox cycle by means of ESI-MS. On the basis of these observations, a mechanism is proposed that may provide an insight into the newly developed aerobic alcohol oxidation.

Imidazolium ionic liquids as solvents for cerium(IV)-mediated oxidation reactions
Use of imidazolium ionic liquids as solvents for organic transformations with tetravalent cerium salts as oxidizing agents was evaluated. Good solubility was found for ammonium hexanitratocerate(IV) (ceric ammonium nitrate, CAN) and cerium(IV) triflate in 1-alkyl-3-methylimidazolium triflate ionic liquids. Oxidation of benzyl alcohol to benzaldehyde in 1-ethyl-3-methylimidazolium triflate was studied by in-situ FTIR spectroscopy and 13C NMR spectroscopy on carbon-13-labeled benzyl alcohol. Careful control of the reaction conditions is necessary because ammonium hexanitratocerate(IV) dissolved in an ionic liquid can transform benzyl alcohol not only into benzaldehyde but also into benzyl nitrate or benzoic acid. The selectivity of the reaction of cerium(IV) triflate with benzyl alcohol in dry ionic liquids depends on the degree of hydration of cerium(IV) triflate: anhydrous cerium(IV) triflate transforms benzyl alcohol into dibenzyl ether, whereas hydrated cerium(IV) triflate affords benzaldehyde as the main reaction product. Reactions of ammonium hexanitratocerate(IV) with organic substrates other than benzyl alcohol have been explored. 1,4-Hydroquinone is quantitatively transformed into 1,4-quinone. Anisole and naphthalene are nitrated. For the cerium-mediated oxidation reactions in ionic liquids, high reaction temperatures are an advantage because under these conditions smaller amounts of byproducts are formed.

The process for producing a (fluoroalkyl)benzene derivative according to the present invention comprises a step of reducing the total content of group 3 to group 12 transition metals in an alkylbenzene derivative to 500 ppm or less in terms of metal atoms; a step of halogenating the branched alkyl group of the purified alkylbenzene derivative by a photohalogenation to obtain a (haloalkyl)benzene derivative; and a step of subjecting the (haloalkyl)benzene derivative to a halogen-fluorine exchange using HF in an amount of 10 mol or higher per one mole of the (haloalkyl)benzene derivative. The (fluoroalkyl)benzene derivative produced by the process is reduced in the content of impurities such as residual halogens and residual metals, and is useful as intermediates for functional chemical products for use in applications such as medicines and electronic materials.

The invention relates to a method for producing aryllithium compounds and to their reaction with suitable electrophiles to obtain compounds of formula (V), by reacting halogen aromatics (I) with lithium metal to obtain an aromatic lithium compound (II), which, as a lithiation agent, reacts with aromatic halogen compounds (III) in a halogen-metal exchange reaction to form the corresponding lithium aromatics (IV). Said aromatics can be reacted in an additional step with a corresponding electrophile to form the desired product (V). The present invention relates to a process for producing a trifluoromethylbenzylamine represented by the general formula (1). This process includes the step of reducing an oxime represented by the general formula (2), where R1represents hydrogen atom, a halogen atom selected from the group consisting of fluorine, chlorine, bromine and iodine, or trifluoromethyl group, where R1is defined as above, and R2represents hydrogen atom, an alkyl group or an aralkyl group. With this process, the trifluoromethylbenzylamine can be produced with high selectivity.

Stereomutation of Methoxycarbenium Ions. 2. Experimental Evidence for an Inversion Process
The stereomutation about the C-O partial double bond of a series of 1-methoxy- (2/3) and 1-hydroxy-3-arylallyl cations (5) has been examined experimentally in strong acid solutions and theoretically by using HF/STO-3G calculations.The interconversions of cations 2/3 were shown to be intramolecular processes in FSO3H, CF3SO3H, and FSO3H/SbF5.The rate constants of the stereomutation of these cations, which were measured by using dinamic NMR methods, were found to depend on the substituents on the aryl ring.Apart from the case where the aryl ring bears two m-CF3 groups, 2g/3g, the rate constants for stereomutation are linearly related to ?+ (ρ = -2.37) with ? donors accelerating the reaction.The calculations performed for 5 show that the lowest energy pathway for isomerization involves rotation rather than inversion.They also reveal that ?-donor substituents on the aryl ring lower the barrier to rotation while raising the barrier to inversion.In both cases, the calculated barriers for substituted systems show a linear correlation with ?+.It is concluded that the preferred pathway for stereomutation of most of these 1-hydroxy and 1-methoxyallyl cations in both strong acid solution and the gas phase involves rotation.When the aryl ring bears very strongly electron-withdrawing groups, e.g., two CF3 groups in 2g/3g, the inversion pathway would seem to be preferred.This is the first example of methoxycarbenium ion which undergoes stereomutation by inversion rather than rotation
