The iodination of 4-amino-2,6-dimethyl-pyrimidine 1a with I2/H2SO4/H2O/H2O2 does not give, as supposed hitherto, the 5-iodo derivative (2) but affords the corresponding 2-iodomethyl derivative 3a.The reaction pathway of the iodination is discussed.The reaction of 3a with nucleophiles leads to a variety of side chain-substituted pyrimidines 5a-j.
