Resolved 2-arylcyclohexylamines: effects on reserpine akinesia and central nervous system monoamine synthesis

A series of cis- and trans-2-arylcyclohexylamines were synthesized and resolved.The absolute configuration was determined by single-crystal X-ray analysis.Some of the compounds, and especially racemic 2-(3-hydroxyphenyl)-N,N-dipropylcyclohexylamine showed a marked locomotor stimulation in reserpinized rats.However, these compounds displayed only minor effects on dopamine and 5-hydroxytryptamine synthesis rates, indicating a non-dopaminergic and/or serotonergic mechanism for this stimulation.
