Rhodamine dyes are utilized effectively as a chemosensor to monitor biologically important metal ions and effectively applied for fluorescent imaging studies in living systems. Herein, rhodamine-B armed fluorescent chemosensor (RhBNC) was designed and synthesized by condensation reaction between rhodamine B hydrazide and naphthyl chromone with ONO binding site in good yield. Upon sensing study towards metal ions, the probe showed selective turn-on fluorescent change for Fe3+ over other cations even in the presence of competing trivalent metal ions as well. The fluorescence switch is “on” when Fe3+ ions binds to RhBNC with orange fluorescence. The probe can detect lower concentration of Fe3+ ions with detection limit of 0.16 μM. Due to its lower detection limit; it can be applied to monitor Fe3+ ion presence in a living system by fluorescent imaging technique. The probe was employed effectively to ascertain the presence of Fe3+ and to image Fe3+ ions in HeLa cell line and zebrafish.

A series of half-sandwich platinum group metal complexes containing coumarin-N-acylhydrazone ligands have been prepared. The metal precursors of the type [(p-cymene)RuCl2]2 and [Cp*MCl2]2 (M = Rh/Ir) and coumarin-N-acylhydrazone ligands (L1, L2 and L3) were reacted in the ratio of 1:2 (M:L), forming neutral bidentate (N ∩ O) complexes (1–9). The complexes are of the general formula [(arene)M{κ2(N∩O)L}Cl]. All these complexes have been characterized by analytical, spectroscopic and single-crystal X-ray diffraction studies. The complexes and ligands were then carried out for antibacterial, antioxidant and DNA binding studies. The results show that both ligands and complexes possess potent antibacterial and antioxidant properties. ___________________________________________________________________________

An efficient one-pot synthesis of alkyl 4-oxo-coumarinyl ethylidene hydrazono-thiazolidin-5-ylidene acetate derivatives has successfully been achieved via a three component cyclization reaction of various substituted 3-acetyl coumarins, thiosemicarbazide and dialkyl acetylenedicarboxylates, in presence of acetic acid. The isolated products were obtained in pure form with high yields through simple workup. The newly synthesised compounds structure was established on the basis of spectral (IR, 1H NMR, 13C NMR, ESI- mass) elemental analysis and single crystal X-ray data. All synthesised compounds were screened for their antiviral activity against a broad spectrum of human viruses in different cell cultures. Of the novel synthesised compounds, thirteen compounds exerted activity against Punta Toro virus, including compound IV-19, for which an antiviral potency was noted against a broad panel of DNA and RNA viruses as well.

Coumarin hydrazone derivatives (L1, L2, L3, and L4) have been prepared by Knoevenagel condensation. Half-sandwich ruthenium, rhodium, and iridium complexes containing these coumarin hydrazone derivatives (1–12) have been synthesized. The deprotonation of hydrazinic proton from the ligand leads to the formation of anionic species, thereby forming neutral N?∩?N chelation complexes. These complexes have been confirmed by spectroscopic technique and single-crystal X-ray diffraction studies. Spectroscopic analysis of the ligands and complexes shows that the data are consistent with the formulated data. The stability studies of some of the complexes (1, 5, and 6) were performed by the 1H-NMR technique using deuterated DMSO-d6 solvent. They are found to be stable in solution as well as in the air and are nonhygroscopic. In vitro antibacterial activity against gram-positive Staphylococcus aureus and gram-negative Escherichia coli, Klebsiella pneumoniae strains have been tested. Also, their cytotoxicity study was evaluated against Dalton's ascites lymphoma (DL), Ehrlich ascites carcinoma (EAC), and normal cell lines (peripheral blood mononuclear cells [PBMC]). In both the studies, complexes (5 and 6) are found to be the most active compared to the other complexes.
