1. The description of bortezomib ( Velcade )
Bortezomib (CAS No. 179324-69-7) is a reversible inhibitor of the chymotrypsin-like activity of the 26S proteasome in mammalian cells. This drug is thought to inhibit many proteins (known as proteasomes) that cancer cells need to survive and multiply. It has been shown to have anti-tumor activity in B cell malignancies.
Bortezomib binds to the proteasome and does so "reversibly". Normal healthy (non-cancer) cells are not as prone to damage from this binding after the Bortezomib comes off, as cancer cells are. Malignant cells suffer a breakdown even after inhibition of the proteasome for a short time.
2. Examples of bortezomib's application
Hangzhou Holypharm biotech Co.,Ltd., a professional supplier of active pharmaceutical ingredients and intermediates, has rich experience on a wide range of chemical reaction and professional research team. Their researchers shows some examples of bortezomib's application in proteasome.
For example, in order to detect the Bortezomib's function to p21 protein, we can process PC - 3 cell with different concentrations of bortezomib (0.5, 1.0, 5.0,,50,100,500 10 nM) and find the p21 protein's concentration in untreated cells is very low. But after the processing of bortezomib in 10 nM, p21 protein's concentration increased significantly. Therefore, the bortezomib is very important in chemical research.
